- Research Interest Statement: Despite significant innovations within the immunotherapy space, metastatic melanoma continues to be difficult to treat, with 50% of patients not receiving clinical benefit with current standard-of-care immune checkpoint inhibitors (ICI). Additionally, of those patients that respond to ICIs, roughly 20-50% of patients develop resistance to the therapy and experience disease progression. Adjuvant or neo-adjuvant therapies are desperately needed to improve overall response rates to ICIs and increase patient disease-free survival. Using clinically relevant Yale University Mouse Melanoma (YUMM) models that have varying degrees of ICI responsiveness, I am analyzing the epigenome and proteome of ICI responsive and innately resistant cell types to evaluate potential targets that could improve patient outcomes on ICIs. In conjunction with innate resistance, I am also developing a model of acquired resistance to ICIs which will allow me to evaluate shared targets between innate and acquired resistance phenotypes. These targets will be used to develop adjuvant therapies that potentially have a larger window of therapeutic benefit for metastatic melanoma patients.
